Hmm. That Dirty Harry “Do ya, punk?” line didn’t precede a kill unless I’m mistaken.
Actually it did. The nutcase decided to try his luck and got blown into a lake.

Al, he’s used it more than once, with very slight variation.
The one i’m quoting the guy wasn’t killed.
But he used the punk line and killed a guy in a later movie.
You could have a long list of excellent lines prior to killing James Bond for the 4,377th time…
In the Dirty Harry movies he always said the key line twice. The first near the beginning of the movie, in an unrelated context, and the second at the climax.
In Dirty Harry the first time was at the scene of a robbery gone bad. He had his .44 pointed at the remaining bad guy, who had a gun in his hand. Harry said ‘I can’t remember whether I fired 6 shots or only 5. Do you feel lucky?’ After the bad guy put down his gun he said ‘I gots to know!’ Harry pulled the trigger. Click.
I think the award for best line and best delivered line goes to The Duke as Rooster Cogburn, “Fill your hand, you sonofabitch!”

Obviously, my 7th decade memory was able to dredge up only the first scene after the robbery gone bad.
A not so obvious immediate kill scene, but my personal favorite nonetheless…Give me back my shoes! (The Bad Seed: Rhoda to Leroy who then proceeds to burn him to death in the maintenance shed)

I think the he remaining bad guy with 12 ga shotgun nearby at the beginning of Dirty Harry was Samuel Jackson (I think maybe one of his first movies). The psychopath got blasted at the end after that line.
